The incorporation of nucleotides into amino acid transfer ribonucleic acid. 3. Thincorporaton of uridylic acid.

چکیده
An enzyme preparation from rabbit muscle catalyzes the incorporation of uridine monophosphate into the terminal nucleotide sequence of yeast amino acid transfer ribonucleic acid. Reactants are UTP, Mg++, and transfer RNA which has been partially digested with snake venom phosphodiesterase. Some characteristics of the incorporation system are presented. Incorporation of UMP into tranfer RNA rather than into a different species of RNA was shown by a similar chromatographic profile of the RNA fraction labeled with UMP, or with @dine monophosphate, by competition between UMP and CMP for a limited number of sites in the RNA molecule, and by nearest neighbor analysis. The latter indicated that 2 UMP residues cannot be incorporated as neighbors in a manner similar to the subterminal CMP residues in transfer RNA.

Effects of Cycloheximide upon Formation of Ribonucleic Acid Cytidylic and Uridylic Acids.
Concentrations of cycloheximide as low as 3 mug/ml inhibited incorporation of labeled orotic acid or uridine into RNA cytidylic acid of soybean (Glycine max) hypocotyl sections. Even lower concentrations of this well known protein synthesis inhibitor interfered with conversion of labeled cytidine into RNA uridylic acid. Both cycloheximide and puromycin inhibited absorption of (3)H-phenylalanine...
متن کاملThe incorporation of nucleotides into amino acid transfer ribonucleic acid. II. Evidence for separate enzymatic sites for incorporation of adenosine 5'-monophosphate and cytidine 5'-monophosphate.
The preceding paper described the purification and some of the properties of an enzyme present in rabbit muscle extracts that catalyzes the incorporation of adenylic acid from adenosine triphosphate into the terminal position of amino acid transfer ribonucleic acid obtained from yeast (1). The data presented in this paper are concerned with the incorporation of cytidine 5’-monophosphate into th...
متن کاملRibonucleic acid code words in wheat germ.
An amino acid-incorporating system has been obtained from ungerminated wheat germ, which can be stimulated by the addition of uridylic-rich polynucleotides. Polyadenylic and polycytidylic acids fail to stimulate the incorporation of lysine and proline, respectively. The amino acid codons determined by the use of the uridylic-rich polynucleotides coincide almost perfectly with those reported for...
متن کاملThe incorporation of adenine nucleotides into ribonucleic acid of cell-free systems from liver.
The isolation and purification of enzymes from bacteria that polymerize ribonucleoside diphosphates (l-3) have intensified the search for enzymes in animals that will synthesize RNA.l A number of reports (4-11) have established that radioactive compounds can be incorporated into RNA in cell-free systems from animals, but the pathway of incorporation and the identity of the immediate precursor o...
متن کاملMetabolism of cytidine and uridine in bean leaves.
The metabolism of cytidine-2-(14)C and uridine-2-(14)C was studied in discs cut from leaflets of bean plants (Phaseolus vulgaris L.). Cytidine was degraded to carbon dioxide and incorporated into RNA at about the same rates as was uridine. Both nucleosides were converted into the same soluble nucleotides, principally uridine diphosphate glucose, suggesting that cytidine was rapidly deaminated t...
